Introduction

Surface plasmon resonance (SPR) is a biophysical technique developed for the detection of molecular interactions based on the principles of physical optics, which allows for the detection of intermolecular interactions in real-time with high sensitivity and without the need for labeling. SPR has been recognized as one of the most powerful techniques for determining the specificity, affinity, and kinetic parameters of a wide range of molecular binding processes of many types, including protein-protein, protein-DNA, enzyme-substrate or inhibitor, etc. The technology can be used for protein interaction studies, small and large molecule drug discovery, antibody screening, and precise characterization, which has been widely used in life sciences, medical diagnostics, food safety, and environmental testing.

Working Principle

SPR technology generates different resonance angles according to whether the ligand coupled to the metal surface binds to the analyte or not. By monitoring the change of the resonance angle, the affinity, affinity constants, binding kinetics, and other information of biomolecular interactions can be inferred.
