Microscale thermophoresis (MST) is an emerging biomolecular interaction analysis technique developed based on the thermophoretic effect, which measures the affinity between a ligand and a target molecule by observing the changes in the conformational size, charge, and solvation state of the target molecule after the ligand binds to it. Currently, the application of MST in life science fields such as drug screening, signal transduction, molecular detection, and medical diagnostics is developing rapidly, which plays an important role in promoting basic biomedical research and better drug development.

Working Principle

MST technology is based on the detection of heat changes caused by interactions between biomolecules. The thermophoretic phenomenon of biomolecules in the capillary will lead to changes in the hydration layer, molecular size, charge, and other molecular properties of the interacting biomolecules, which in turn causes changes in the fluorescence distribution in the reaction system to detect the affinity between the interacting biomolecules.


  • Measurements can be accomplished in a variety of different solution environments, including the complex detergent environments required to study membrane proteins.

  • Samples do not need to be immobilized and measurements in biological solutions do not require purification.
